Advancedtechniquesincludingthechromosomeconformationcapture(3C)methodologyanditsderivativesarecomplementingmicroscopyapproachestostudygenomeorganization,andarerevealingnewdetailsofthree‐dimensional(3D)genomearchitectureatincreasingresolution.ThefissionyeastSchizosaccharomycespombe(S.pombe)comprisesasmallgenomefeaturingorganizationalelementsofmorecomplexeukaryoticsystems,includingconservedheterochromatinassemblymachinery.Herewereviewkeyinsightsintogenomeorganizationrevealedinthismodelsystemthroughavarietyoftechniques.WediscussthepredominantroleofRabl‐likeconfigurationforinterphasechromosomeorganizationandthedynamicchangesthatoccurduringmitosisandmeiosis.HighresolutionHi‐Cstudieshavealsorevealedthepresenceoflocallycrumpledchromatinregionscalled“globules”alongchromosomearms,andimplicatedacriticalroleforpericentromericheterochromatininimposingfundamentalconstraintsonthegenometomaintainchromosometerritorialityandstability.Thesefindingshaveshednewlightontheconnectionsbetweengenomeorganizationandfunction.ItislikelythatinsightsgainedfromtheS.pombesystemwillalsobroadlyapplytohighereukaryotes...
