Conjugativeplasmidtransferresultsinthespreadofantibioticresistancegenesandvirulencefactorsbetweenbacterialcells.PlasmidtransferisdependentupontheDNAnickingactivityofaplasmid‐encodedrelaxaseenzyme.TyrosineresidueswithintherelaxasecleavetheDNAplasmidnicsiteinahighlysequence‐specificmanner.TheconjugativeresistanceplasmidpCU1encodesarelaxasewithfourtyrosineresiduessurroundingitsactivesite(Y18,19,26,27).WeuseactivityassaystodemonstratethatthepCU1relaxasepreferentiallyusesY26oracombinationofY18+19tonickDNAatwildtypelevels,andthatanadjacentasparticaciddeprotonatesthesetyrosinestoactivatethemforattack.OurfindingsillustratetheuniquemodificationsthatthepCU1relaxasehasintroducedintothetraditionalrelaxase‐mediatedDNAnickingmechanism...
