Proteinsplicingcatalyzedbyinteinshasenabledvariousbiotechnologicalapplicationssuchasproteinligation.Successfulapplicationsofinteinsareoftenlimitedbysplicingefficiency.Here,wereportthecomparisonofproteinsplicingbetween20differentinteinsfromvariousorganismsinidenticalcontextstoidentifyrobustinteinswithforeignexteins.WefoundthatRadAinteinfromPyrococcushorikoshiiandanengineeredDnaBinteinfromNostocpunctiformedemonstratedanequallyefficientsplicingactivitytothepreviouslyreportedhighlyefficientDnaEinteinfromNostocpunctiforme.Thenewlyidentifiedinteinswithefficientcis‐splicingactivitycanbegoodstartingpointsforthefurtherdevelopmentofnewproteinengineeringtools...
