Localdrugdeliveryhasreceivedmuchrecognitioninrecentyears,yetitisstillunpredictablehowdrugefficacydependsonphysicochemicalpropertiesanddeliverykinetics.Thepurposeofthecurrentstudyistoprovideausefulmathematicalmodelfordrugreleasefromadrugdeliverydeviceandconsecutivedrugtransportinbiologicaltissue,therebyaidingthedevelopmentofnewtherapeuticdrugbyasystemicapproach.Inordertostudythecompleteprocess,atwo-layerspatio-temporalmodeldepictingdrugtransportbetweenthecoupledmediaispresented.Drugreleaseisdescribedbyconsideringsolubilisationdynamicsofdrugparticle,diffusionofthesolubiliseddrugthroughporousmatrixandalsosomeotherprocesseslikereversibledissociation/recrystallization,drugparticle-receptorbindingandinternalizationphenomena.Themodelhasledtoasystemofpartialdifferentialequationsdescribingtheimportantpropertiesofdrugkinetics.Thismodelcontributestowardstheperceptionoftherolesplayedbydiffusion,mass-transfer,particlebindingandinternalizationparameters...
