用蚕丝丝素(SF)和聚己内酯(PCL)共混进行静电纺丝,可以制备一种新的既具有良好生物相容性又具有良好力学性能的可降解性生物材料。用激光共聚焦显微镜、扫描电镜、细胞粘附率和四甲基偶氮唑盐比色法对SF/PCL共混纤维的细胞相容性进行检测和分析,研究了SF/PCL共混比例对L-929细胞粘附和生长的影响。结果表明,细胞在SF/PCL静电纺丝网表面能够正常粘附和增殖且形态正常,随着培养时间的延长细胞数量逐渐增多,当SF/PCL共混比例为70/30和50/50时,细胞粘附率和增殖率明显高于其他比例的静电纺丝网。>In order to develop a new degradable biomaterial with both good biocompatibility and mechanical pro- perties, blended fibers composed of silk fibroin (SF) and poly(ε-caprolactone) (PCL) were fabricated by electrospining technique in this study. Confocal laser scanning microscope, scanning electron microscopy, cell adhesion rate and MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay were used to analyse the cytocompatibility of SF/PCL electrospun fibers, and to study the influence of the SF/PCL blend ratio on L-929 cell adhere and proliferate. The results showed that cells on the SF/PCL electrospun fibers surface can adhere and proliferate, and the cell number increased with the prolongation of the culturing time. The rate of cell adhesion and proliferation cultured on the SF/PCL electrospun fibers with the ratio of 70/30 and 50/50 was higher than other SF/PCL blend ratio electrospun fibers.
