Methods of culturing in vitro an African endemic species of Nicotiana africana (Solanaceae) possessing a number of valuable economic features are developed. The influence of different concentrations of auxin (naphthyleneacetic acid) and cytokinins (benzylaminopurine, zeatin, thidiazuron) on morphogenesis and regeneration of N. africana plants with the use of shoots and internodes as explants is analyzed. The optimum method of regeneration of N. africana plants in explants of shoots is by culturing in a medium containing 0.1 mg/1 α-naphthaleneacetic acid and 1 mg/l 6-benzylaminopurine, and in explants of internodes, in a medium containing 0.5 mg/l α-naphthaleneacetic acid and 1 mg/l zeatin. The use of thidiazuron proved to be most effective at a concentration of 0.4 mg/l followed by transplanting of the explants into a hormone-free medium. A technique of isolating and culturing mesophyll protoplasts that makes possible regeneration of N. africana plants in 3-4 months is proposed.
