Evaluation of Chemiluminescence Immunoassays for Detecting Thyroglobulin (TG) and Thyroid Peroxidase (TPO) Autoantibodies Using the IMMULITE~® System

The chemiluminescence immunoassays for detecting autoantibodies to thyroid peroxidase (TPO) and thyroglobulin (TG) were evaluated using the IMMULITE~® immunoanalyzer supplied by Diagnostics Products Corporation (DPC). These were immunometric (sandwich) assays with antigen-coated beads. There was good reproducibility of the measured results with an intra-assay precision of 2.3 - 3.9% for anti-TG and of 3.5 - 5.6% for anti-TPO and an inter-assay precision of 7.1 - 7.5% for anti-TG and 7.8 - 10.5% for anti-TPO. The lower detection limit was determined as 20 IU/ml for anti-TG and 7 IU/ml for anti-TPO. Dilution linearity, with an average of 103% for anti-TPO and 101% for anti-TG, and average recovery rates of 93% for anti-TPO and 94% for anti-TG, were acceptable. The clinical findings of the tests were compared with test systems from ORGenTec, ELIAS Medizintechnik, Boehringer Mannheim and B.R.A.H.M.S. Clinical concordance taking the respective cutoff values into account was 92% - 96% for anti-TG and 93% and 98% for anti-TPO. Further samples from patients with clinically diagnosed Hashimoto's thyroiditis were investigated. The levels of positive anti-TG values (83%) and anti-TPO values (94%) accorded with those quoted in the literature. In den untersuchen Ringversuchsproben von INSTAND e.V. wurde eine gute Übereinstimmung mit den erwarteten klinischen Ergebnissen erzielt.
