Molecular imaging of separate but still incompletelynresolved spots on high-performance thin-layer chromatographyn(HPTLC) plates is used for the direct analysisnof porcine brain lipids by desorption electrospray ionizationnmass spectrometry (DESI-MS). Eight class-specificnspots were imaged in the negative ion mode and shownnto contain more than fifty lipids. A low lateral resolutionnof 400 × 400 μm allowed simple, rapid, and incompletenseparation to be combined with DESI imaging for thenidentification of many components of these extremelyncomplex mixtures. In this work, tandem mass spectrometryn(MS/MS) was also employed to confirm the identitynof particular lipids directly on HPTLC plates.
