A dual-functionalized nanoprobe was designed for highlynsensitive and selective in situ evaluation of carbohydratesnon living cells by integrating the specific carbohydratenrecognition and enzymatic signal amplification of proteinsnon Au nanoparticles. A nanoscaffold of nanohorns functionalizednwith arginine-glycine-aspartic acid-serine tetrapeptidenwas also prepared on an electrode surface forncell capture and enhancing the electrical connectivity.nCombined with the nanoprobe and peptide-nanohorns, anhighly sensitive electrochemical strategy was developednfor cytosensing, which showed a detection limit down ton15 cells, broad dynamic range, acceptable rapidity, andnlow cost. The proposed method was further used fornmonitoring of dynamic variation of carbohydrate expressionnon cancer cells in response to drugs, which obviatednthe destruction or labeling of cells and the covalent taggingnof lectin and enzyme. The one-pot conjugation of threencomponents was very convenient and could be extendednfor preparation of other lectin-functionalized nanoprobes.nFurther development of this technique would contributenconsiderably to meeting the challenges in comprehensivenunderstanding of the glycomic codes.
