Increased Oxidative Modifications of Amniotic Fluid Albumin in Pregnancies Associated with Gestational Diabetes Mellitus

摘要

Gestational diabetes mellitus (GDM) is a state of hyperglycaemianand increased oxidative stress with onset duringnpregnancy. Human serum albumin (HSA) was extractednfrom 26 GDM and 26 nonGDM amniotic fluidnsamples collected at 15 weeks gestation and analyzed bynmass spectrometry. The majority of all albumin isoformsnwere oxidized with the cysteinylated HSA as the base peaknin the deconvoluted spectrum. The HSA peak areas, fromna control sample, had 36% relative standard deviationn(RSD) across the six experimental days but using thenrelative isoform distribution improved the precision ton3-6%. The results show that the relative contribution ofnpermanently oxidized HSA was greater (P ) 0.002) andnreversibly oxidized HSA was lower (P ) 0.006) for GDMncompared to nonGDM in the samples measured. Thisnimplies that the path toward GDM has been set prior ton15 weeks gestation and results in increased proteinnoxidation.
