花椒多酚类化合物对脑卒中后抑郁大鼠脑组织NE、5-HT及MAO的影响

         

摘要

Objective To explore the mechanism of polyphenol extract from Zanthoxylum bungeanum Maxim. (ZPPC) on treating post-stroke depression. Methods Post-stroke depression model of rat was established. ELISA was used to detect the influence of ZPPC and fluoxetine on norepinephrine (NE) and 5-hydroxytryptamine (5-HT) contents in brain tissue and the monoamine oxidase (MAO) activity in rats brain was determined by fluorescence spectrophotometry. Results The levels of NE and 5-HT in brain tissue was significantly lower in model group as compared with control group (P<0.05), but dramatically elevated after treatment with high-dose ZPPC(200mg/kg) and fluoxetine (P<0.05). The activity of MAO in rats brain was inhibited by intervention of ZPPC with 100 and 200 mg/kg as compared with model group(P<0.05). Conclusion ZPPC can upregulate NE and 5-HT in brain tis-sue of rats with post-stroke depression and the underlying mechanism may involve inhibition of MAO activity.%目的:探讨花椒多酚类化合物总提取物(ZPPC)抗脑卒中后抑郁作用机制。方法建立脑卒中后抑郁大鼠模型,酶联免疫法检测ZPPC、氟西汀对受试大鼠脑组织去甲肾上腺素(NE)、5-羟色胺(5-HT)含量的影响;采用荧光分光光度法测定ZPPC和吗氯贝胺对受试大鼠脑内单胺氧化酶(MAO)活性的影响。结果与空白对照组比较,模型组NE、5-HT水平显著降低(P约0.05),ZPPC高剂量组(200mg/kg)和氟西汀组与模型组比较,NE、5-HT水平均有显著性升高(P约0.05);ZPPC中、高剂量组(100mg/kg和200mg/kg)还明显抑制大鼠脑组织MAO-A活性(P约0.05)。结论 ZPPC组能显著提高脑卒中后抑郁大鼠脑组织NE、5-HT水平,其作用机制可能与抑制脑组织MAO-A活性有关。
