夏枯草口服液对STAT3和NF-KB信号转导通路的影响

             

摘要

目的 探讨夏枯草口服液对炎症相关STAT3和NF-KB信号通路的抑制作用.方法 应用稳定转染STAT1-luciferase和STAT3-luciferase报告基因质粒的HepG2细胞及稳定转染NF-KB-luciferase报告基因质粒的293细胞,分别研究夏枯草口服液对STAT3和NF-KB信号通路的影响.结果 夏枯草口服液10 mg/ml、5mg/ml、1mg/ml、0.5mg/ml和0.1mg/ml对STAT3信号通路抑制率分别为98.39%、96.77%、81.53%、57.18%和15.73%,IC50为0.4591mg/ml,对NF-KB信号通路抑制率分别为99.92%、99.48%、91.29%、58.36%和-11.30%,IC50为0.3719 mg/ml.结论 夏枯草口服液抗炎作用的机制可能与抑制STAT3和NF-KB信号通路有关.%Objective To investigate the inhibition effects of prunella vulgaris oral liquid on inflammatory related STAT3 and NF-KB signaling pathways. Method The effects of prunella vulgaris oral liquid on the STAT3 and NF-KB signaling pathways were studied by stable transfection of STAT1-luciferase and STAT3-luciferase reporter gene plasmid HepG2 cells and 293 cells stably transfected with NF-KB-luciferase reporter gene plasmid. Result The inhibitory rates of 10mg/ml,5mg/ml,1mg/ml,0.5mg/ml and 0.1mg/ml of prunella vulgaris oral liquid were 98.39%,96.77%, 81.53%,57.18% and 15.73% respectively,and IC50 was 0.4591mg/ml,and the inhibition rates for NF-KB signal pathways were 99.92%,99.48%, 91.29%,58.36% and-11.30% respectively,the IC50 was 0.3719mg/ml. Conclusion The prunella vulgaris oral liquid may play an anti-inflammatory role by inhibiting STAT3 and NF-KB signaling pathway.
