目的 观察佛波酯(12-myristate 13-acetate,PMA)不同浓度和不同时间对人体胃癌BGC823细胞MMP-9表达的影响,探讨PMA能诱导MMP-9表达增加的最佳浓度和诱导时间.方法 采用mRNA(Real Time RT-PCR)蛋白(West-blot)明胶酶活性分析(Zymography)观察不同浓度的PMA作用于人胃癌BGC823细胞不同时间后的MMP-9表达.结果 PMA浓度1 5mg/L、3 0mg/L、6 0mg/L、12 0mg/L、24 0mg/L 24h时对人胃癌BGC823细胞作用后MMP-9表达率分别为(19 1±1 27)%、(25 3±5 7)%、(52 6±4 8)%、(67 9±3 6)%、(91 3±2 3)%;48h时分别为(21 1±1 9)%、(29 2±3 6)%、(56 6±4 3)%、(76 1±5 1)%、(93 3±2 9)%;72h时分别为(23 4±3 7)%、(37 8±4 2)%、(60 3±14 5)%、(83 6±18 1)%、(95 6±1 3)%.结论 PMA对MMP-9表达的影响浓度剂量和时间依赖性,最佳浓度24 0mg/L,最佳时间24h.%Objective To observe the expression of MMP - 9 in Human gastric cancer BGC823 at different density of PMA of different time, to investigate the optimizated time and density of PMA induce expression of MMP -9. Results The density of PMA was 1. 5mg/L, 3.0mg/L, 6.0mg/L, 12.0mg/L, 24.0mg/L, after 24h on BGC823 cells, the MMP - 9's expression was(19. 1 ± 1. 27)% , (25. 3 ± 5. 7)% , (52. 6 ±4.8)% , (67. 9 ± 3. 6) %、 (91. 3 ± 2. 3)%;affter 48h, it was(21. 1 ± 1. 9) % 、(29. 2 ±3. 6) % 、(56. 6 ±4. 3) % ,(76. 1 ±5.1)%、(93. 3 ±2. 9) % ;after 72h it was (23. 4 ± 3. 7) % , ( 37. 8 ± 4. 2) % , (60. 3 ± 14. 5 ) % , ( 83. 6 ± 18.1) % , (95. 6 ± 1. 3 ) % . Conclusion PMA induces The expression of MMP-9, which is dentisy and time dependent.
