叔丁基对二苯酚对蛛网膜下腔出血后早期脑损伤的保护作用

             

摘要

目的:探讨叔丁基对二苯酚(tBHQ)对蛛网膜下腔出血(SAH)所致的早期脑损伤的保护作用。方法40只正常成年健康雄性SD大鼠,随机分为对照组,SAH组,安慰剂组以及tBHQ处理组,经大鼠自体尾动脉或股动脉非肝素化动脉血注入视交叉池致蛛网膜下腔出血模型。tBHQ通过灌胃给药,剂量为12.5mg/kg,分别在SAH后2h、12h、36h给药,在蛛网膜下腔出血48h后处死。取大鼠血凝块周围的皮层脑组织(额颞底)做标本,测定水肿指数,凋亡及坏死指数。结果与对照组相比,SAH组及安慰剂组的脑水肿指数、凋亡及坏死指数明显增高(P<0.05),而SAH组与安慰剂组差异无统计学意义(P>0.05)。且tBHQ组水平较对照组SAH组和安慰剂组的水肿指数、凋亡和坏死指数则显著下调(P<0.05)。结论 tBHQ对蛛网膜下腔出血后的早期脑损伤具有保护作用。%Objective TO investigate the protective effect of tert-butyl hydroquinone(tBHQ)on early brain injury after subarachnoid hemorrare in rat subarachnoid hemorrhage (SAH) model. Methods Forty male Sprague Dawley rats were randomly divided into four groups,i. e. control group,SAH group,SAH placebo group and SAH plus tBHQ group. All SAH animals were subjected to injection of 0.3ml fresh arterial non-heparinized blood into prechiasmatic cistern. In SAH tBHQ group,tBHQ was administered via oral gavage at a dose of 12.5mg/kg at 2h,12h,and 36h after SAH. Brian samples were extracted and evaluated at 48 h after SAH. The brain tissue adjacent to the clotted blood was taken to analysis in our study. Water content of the local brain in tissue was measured,and apoptosis cells and necrosis cells were detected by TUNEL and FJB. Rusults The brain edema index, apoptosis cell percentage and necrosis cell percentage were increased significantly following SAH(P<0.05). There was no statistically significant difference between SAH group and SAH tBHQ group (P>0.05). The brain edema index,apoptosis cell percentage and necrosis cell percentage were siginificantly reduced (P<0.05). Conclusion tBHQ administration resulted in abatement of the development of early brain injury in this prechiasmatic cistern SAH model.
