不同浓度聚维酮碘溶液对腹膜组织影响的实验研究

             

摘要

目的:观察不同浓度聚维酮碘(PVP-I)溶液对腹膜组织的影响,探讨用于腹腔冲洗的合适浓度。方法将60只SD大鼠随机分为5组,每组各12只,分别将生理盐水及四种不同浓度的PVP-I溶液(0.1% PVP-I高浓度组、0.05% PVP-I中浓度组、0.025% PVP-I亚中浓度组、0.01% PVP-I低浓度组)2ml对各组大鼠进行腹腔内灌注。在灌注后48h打开大鼠腹腔,观察腹腔内有无腹水产生,并对腹水进行采集计量、测定腹水中的白细胞及红细胞数量,采集腹膜组织行组织学检查,对腹膜组织的厚度及腹膜组织中炎性细胞的浸润数量进行观察计量。结果0.1% PVP-I高浓度组及0.05% PVP-I中浓度组的大鼠腹腔内均有腹水产生,并能在腹水中检测出大量白细胞与红细胞,组织学检查发现该两组的腹膜组织厚度与腹膜炎性细胞的浸润数量均显著高于其余各组(P<0.05)。结论≥0.05%浓度的 PVP-I溶液会导致腹膜组织的炎性反应,外科手术中使用聚维酮碘溶液进行腹腔冲洗时需的浓度应<0.05%。%Objective To investigate the impact of different concentration of povidone-iodine(PVP-I)on peritoneum and to determine the optimal concentration for intraoperative peritoneal irrigation. Methods Intraperitoneal infusion were carried out on 60 SD mice which were divided into 5 groups with 12 for each. 2ml of physiological saline or indicated concentration of PVP-I(0.1%,0.05%,0.025% and 0.01%)were used for infusion. After 48 hours,the ascites were collected and assessed for the amount of leucocyte and erythrocyte. The infiltrated inflammatory cells in peritoneum tissues were also analyzed. Results Ascites were found in those mice which were treated with 0.1% and 0.05% PVP-I. There were also more ascites leucocytes and erythrocytes,as well as tissue infiltrated inflammatory cells,in these two groups. Conclusion PVP-I would injury the normal peritoneal tissues at a high concentration(≥0.05%)which implied that the optimal concentration for intraoperative irrigation should be less than 0.05%.
