欧洲花楸成熟种胚组织培养技术

         

摘要

In order to provide a technical basis for remote introduction and rapid propagation of Sorbus pohuashanensis,using mature embryo in S.pohuashanensis as explants and MS as basic medium,the techniques of adventitious buds introduction,proliferation,rooting and regenation plants formation were studied.The results show that the suitable medium for inducing adventitious buds of mature embryo in S.pohuashanensis is MS supplemented with 6-BA 0.5 mg/L,NAA 0.5 mg/L,sucrose 30 g/L and agar 6 g/L,and its induction rate is 80.33%.This kind of medium can induce calli and adventitious buds with high quality and the larger breeding potiencial.The optimum medium for inducing adventitious buds proliferation is MS supplemented with 6-BA 0.2 mg/L,IAA 0.5 mg/L,sucrose 30 g/L and agar 6 g/L,and it can induces 5-13 adventitious buds.The best medium for rooting is 1/2MS supplemented with IBA 0.2 mg/L,sucrose 30 g/L and agar 6 g/L,and the roofing rate is 70%.%为了给欧洲花楸远距离引种和快繁提供技术依据,以欧洲花楸成熟种胚为外植体,以MS为基本培养基,对其不定芽诱导、增殖、生根等再生植株形成技术进行了研究.结果表明:诱导花楸成熟种胚产生不定芽的适宜培养基为MS+6-BA 0.5 mg/L+NAA 0.5 mg/L+蔗糖30g/L+琼脂6g/L,其成熟种胚不定芽诱导率为80.33%.该培养基上诱导出的愈伤组织和不定芽质量高,具有较大的繁殖潜力.诱导不定芽增殖的最适培养基为MS+6-BA 0.2 mg/L+IAA 0.5 mg/L+蔗糖30 g/L十琼脂6 g/L,可诱导增殖出5~13个不定芽.生根培养效果最好的培养基是1/2MS+IBA 0.2 mg/L+蔗糖30 g/L+琼脂6 g/L,成熟种胚不定芽生根率为70%.
