羌活鱼水提物在大鼠骨髓间充质细胞成骨中的作用

         

摘要

Objective: To explore the function of serum containing aqueous extract from QiangHuoYu on the formation of rats' bone marrow mesenchymal stem cells (BMSCs). Method: Bone marrow mesenchymal cells of 3rd generation were divided into 4 groups. 4 groups were respectively cultured on the media of saline+DMEF/F12 culture solution+osteogenous differentiation inducing liquid+5%FBS (saline control group), high-dose aqueous extract from QiangHuoYu+ DMEF/F12 culture solution+osteogenous differentiation inducing liquid+5%FBS (high-dose aqueous extract group of QiangHuoYu), moderate-dose aqueous extract from ()tang'//uoFu+DMEF/F12 culture solution+osteogenous differentiation inducing liquid+5%FBS (moderate-dose aqueous extract group of QiangHuoYu) and low-dose aqueous extract from QiangHuoYu+ DMEF/F12 culture solution+osteogenous differentiation inducing liquid+5%FBS (low-dose aqueous extract group of Qiang Huo Yu), when the fusion of bone marrow mesenchymal stem cells reached 80%. The degrees of osteogenous differentiation were assessed by the activity of Alpase, sediment yield of calcium salt and the node amounts of calcification.The data was analyzed by using single factor variance analysis in SPSS 16.0 software package. Result: Group of Serum containing aqueous extract from QiangHuoYu showed higher activity of Alpase, higher sediment yield of calcium salt and higher secretion of osteocalcin than saline control group. Conclusion: Serum containing aqueous extract from QiangHuoYu can improve the ability of BMSCs osteogenous differentiation induced by osteogenous differentiation inducing liquid, showing higher activity of Alpase and ability of matrix mineralization, which plays a key role in osteogenous differentiation of BMSCs.%目的:探讨羌活鱼水提物含药血清对大鼠骨髓间充质细胞(BMSCs)成骨的作用.方法:取大鼠第三代BMSCs分为4组,待BMSCs接近80%融合时,分别应用生理盐水+DMEM/F12培养液+成骨分化诱导液+5%FBS(生理盐水对照组)、高剂量羌活鱼水提物+DMEM/F12培养液+成骨分化诱导液+5%FBS(羌活鱼水提物高剂量组)、中剂量羌活鱼水提物+DMBM/F12培养液+成骨分化诱导液+5%FBS(羌活鱼水提物中剂量组)、低刺量羌活鱼水提物+DMEM/F12培养液+成骨分化诱导液+5%FBS(羌活鱼水提物低剂量组)培养基培养.应用碱性磷酸酶(ALP)活性、钙盐沉积量及钙化结节数检测成骨分化程度.采用SPSS 16.0软件包对数据进行单因素方差分析.结果:羌活鱼水提物含药血清组较生理盐水对照组表现出高ALP活性、高钙盐沉积量、高骨钙素分泌量.结论:羌活鱼水提物含药血清可提高成骨性诱导液诱导BMSCs成骨分化的能力,表现出高ALP活性及基质矿化能力,在BMSCs成骨分化中可起到重要作用.
