四神丸对溃疡性结肠炎大鼠结肠黏膜PPAR-γmRNA和蛋白表达的影响

Objective: To observe the effects ofSiShenpills on the expressions of PPAR-γmRNA and protein in colonic mucosa of TNBS/alcohol enema-induced ulcerative colitis(UC) rats, and explore its mechanism. Methods: Sixty rats were randomized into the blank group, the model group,SiShenpills group (5 g/kg ) and SASP group (0.3 g/kg), UC rat model was replicated by TNBS/alcohol enema, gross morphological injuries of the colon were observed under the naked eyes and scored, the expressions of PPAR-γmRNA and protein were detected by RT-PCR and immunohistochemical method. Results: The inflammation and ulceration could be seen in the mucous layer of colon tissue of the rats in the model group; compared with the blank group, the expressions of PPAR-γmRNA and protein decreased notably in the colon tissue of the rats in the model group, and the difference presented statistical meaning (P<0.01); compared with the model group, the expressions of PPAR-γmRNA and protein raised evidently inSiShen pills group, and the difference demonstrated statistical meaning(P<0.01). Conclusion:SiShenpills could up-regulate the expressions of PPAR-γmRNA and protein in colon mucosa of UC rats, which suggests that the mechanism of SiShenpills in treating UC might be related to PPAR-γ-related signal transduction pathway regulation.%目的：观察四神丸对三硝基苯磺酸(trinitrobenzene sulfonic acid，TNBS)/乙醇灌肠所致溃疡性结肠炎(ulcerative colitis，UC)模型大鼠结肠黏膜PPAR-γmRNA和蛋白表达的影响，探讨其作用机制。方法：将Wistar大鼠60只随机分为空白组、模型组、四神丸组(5 g/kg)及柳氮磺吡啶(SASP，0.3 g/kg)组，采用TNBS/乙醇灌肠法复制UC大鼠模型，灌胃给药后肉眼观察结肠大体形态损伤并评分，采用RT-PCR和免疫组化法检测PPAR-γ mRNA和蛋白的表达。结果：模型组大鼠结肠组织黏膜层可见炎症和溃疡形成；与空白组比较，模型组大鼠结肠组织PPAR-γ mRNA和蛋白表达量降低，差异有统计学意义(P＜0.01)；与模型组比较，四神丸组PPAR-γmRNA和蛋白的表达量升高，差异有统计学意义(P＜0.01)。结论：四神丸对TNBS/乙醇法所致UC大鼠模型结肠黏膜PPAR-γ mRNA和蛋白的表达有上调作用，提示四神丸治疗UC的作用机制可能与调控PPAR-γ相关信号转导途径有关。
