Objective To investigate the effects of danshensu on the proliferation, migration and invasion of oral squamous cell carcinoma CAL-27 in vitro. Methods Danshensu was used to treat CAL-27 cells, and its effect on the activity of CAL-27 cells was detected by MTT assay. The scratch test was applied to determine the effect of danshensu on the migration ability of CAL-27 cells. The transwell assay was carried out to inspect its effect on the invasive ability of cells. Results MTT showed that danshensu inhibited the proliferation of CAL-27 cells in a dose-and time-dependent manner under concentrations of 40, 60 and 80 μg/m L. The results of the scratch test showed that the difference between the 24 h group and the control group was statistically significant (P＜0.01). The results of Transwell invasion assay showed that danshensu could reduce the invasive ability of CAL-27 cells. Conclusion In vitro, danshensu can inhibit the proliferation of CAL-27 cells and reduce the migration and invasion of CAL-27 cells in a certain range of concentrations.%目的 探讨丹参素在体外条件下对口腔鳞癌细胞CAL-27的增殖、迁移及侵袭能力的影响.方法 丹参素处理CAL-27细胞,MTT法检测丹参素对CAL-27细胞活性的影响.划痕实验检测丹参素对CAL-27细胞迁移能力的影响.Transwell实验检测其对细胞侵袭能力的影响.结果 MTT法显示,在丹参素浓度为40、60和80μg/m L时,对CAL-27细胞增殖的抑制可呈剂量和时间依赖性.划痕实验结果显示,24 h组与对照组相比,差异具有统计学意义(P＜0.01).Transwell侵袭实验结果表明,丹参素可降低CAL-27细胞的侵袭能力.结论 在体外条件下,丹参素在一定浓度范围内可抑制CAL-27细胞的增殖,降低CAL-27细胞的迁移及侵袭能力.
