不同时期成釉细胞氟损伤程度差异的动物实验研究

摘要

Objective To study the expression of GRP78 and BCL-2 in ameloblasts of fluorosis rats and to explore the injury diversi-ty of fluoride to ameloblasts at different stages.Methods 20 Wistar rats were selected and fed with fluoride water.8 weeks later,they were sacrificed.Immunohistochemistry and HE staining were used to detect the changes of ameloblast and the expression of GRP78, BCL-2 in rat incisors.Metamorphic microscope images analysis system and SPSS13.0 software were used to analyze the data.Results The expression of GRP78 in ameloblast pre-secretory phase and secretory phase was higher than that in transformation period and ma-ture period.BCL-2 had the highest expression in secretory phase and transition period,however,it declined in pre-secretory stage and mature stage.The differences between groups were statistically significant(P <0.05 ).Conclusions The injury of fluoride to amelo-blasts at different times is different.%目的：检测 GRP78和 BCL -2在氟中毒大鼠成釉细胞中的表达和分布，探索不同时期成釉细胞氟损伤程度差异。方法选择20只 Wistar 大鼠，饲喂氟浓度为75 mg ／L 的自来水，8周后处死，通过 HE 染色和免疫组化技术观察大鼠成釉细胞形态，以及 GRP78和 BCL -2在氟中毒大鼠不同时期成釉细胞中的表达。结果GRP78在成釉细胞分泌前期和分泌期表达高于转换期和成熟期，BCL -2在分泌期和转换期表达最高，在分泌前期和成熟期表达下降，各组间具有统计学差异（P ＜0．05）。结论处于不同时期的成釉细胞对氟损伤程度存在差异。
