血小板衍生生长因子受体α基因在C57 BL／6 J小鼠胚胎腭突融合过程中表达变化的研究

Objective To observe the expression of PDGFR-αin the development stages of mouse embryo palatal shelves.Methods Palatal shelves of C57BL/6J inbred mouse embryos were obtained on the day of GD13.5,GD14.5 and GD15.5 respectively by using stereoscope microscope.The expression location of PDGFR-αprotein in the palatal shelf was observed by immunohistochemistry( IHC) staining;and the quantities expression of PDGFR-αprotein were examined by Western blot(WB) analysis.Results IHC staining showed that PDGFR-αwas specifically expressed in mouse palatal shelves.In GD13.5,PDGFR-αexpressed mainly in the palatal epi-thelium;but in GD14.5,GD15.0,and GD15.5,the PDGFR-αexpressed both in mesenchymal cells and epithelial cells.The WB analy-sis showed that proteins expression of PDGFR-αin different developmental stages of palatal was consistent with IHC results.In GD14. 5,the expression level of PDGFR-αreached the peak.The proteins expression in GD13.5,GD15.0 or GD15.5 was significantly differ-ent compared with that of in GD14.5.Conclusions The study showed PDGFR-αhad spatiotemporal expression features during the de-velopment stages of mouse embryo palatal shelves.PDGFR-αgene may be associated with the development of palatal shelves.%目的：观察小鼠胚胎腭突融合过程中血小板衍生生长因子受体α基因（ platelet derived growth factor receptor alpha， PDGFR－α）的表达特点。方法 C57BL／6J近交系受孕小鼠分别于妊娠13．5 d（Gestation Day，GD13．5）、GD14．5、GD15．0、及GD15．5时断颈处死。取小鼠胚头及腭突，免疫组织化学检测不同胚胎发育时期的腭突组织中PDGFR－α蛋白的表达定位；Western blot法检测PDGFR－α蛋白的表达变化。结果免疫组化检测结果显示PDGFR－α在小鼠腭突中特异性表达：在GD13．5时主要表达在腭突上皮，而在GD14．5、GD15．0、GD15．5时在间充质和上皮都有表达，且在GD14．5时PDGFR－α的表达水平达到高峰。 Western blot检测PDGFR－α在腭突不同发育时期的蛋白表达变化和免疫组化检测结果趋势一致。在GD14．5时，PDGFR－α的表达水平达到高峰，和GD13．5、GD15．0及GD15．5蛋白表达水平相比差异有统计学意义。结论研究证实PDGFR－α在小鼠胚胎腭突发育过程中存在时空表达变化的特征。 PDGFR－α可能与胚胎腭突的发生有关。
