石胡荽组织培养及无性系建立的研究

         

摘要

In order to protect wild resources and realize artificial cultivation, the tender stems of Centipeda minima were used as materials to carry out the research on adventitious bud induction and differentiation, rooting and transplanting of tube seedlings and so on. The results showed that the optimum medium for adventitious bud induction was MS+6-BA 0.4mg/L+NAA 0.1mg/L;MS+NAA 0.1mg/L+GA3 0.5mg/L+6-BA 0.6mg/L was the ideal medium for adventitious bud differentiation. After dealing with IAA (concentration of 40mg/L) for 6 minutes, the adventitious buds could be easily induced rooting in the medium of Ng+NAA 0.2mg/L. The tube seedlings could be easy to survive after transplanting. Colonization of plantlets maintained all the biological traits which wild plants have. Finally the clone of Centipeda minima was established.%为了保护野生资源,实现石胡荽人工栽培,我们以石胡荽嫩茎为材料进行了不定芽诱导与分化、试管苗生根、继代、移栽和定植研究.结果表明:MS+6-BA 0.4mg/L+NAA 0.1mg/L是石胡荽嫩茎直接诱导分化不定芽的理想培养基;MS+NAA0.1 mg/L+GA3 0.5mg/L+6-BA 0.6mg/L是石胡荽不定芽增殖分化培养的理想培养基;把不定芽下部切口浸入浓度为40mg/L的IAA溶液处理6min后,接种到N6+NAA0.2mg/L培养基上进行生根培养的方法是石胡荽不定芽生根培养的理想方法.试管苗的移栽和定植易成活,定植的试管苗保持了野生石胡荽的所有植物学性状.建立起石胡荽嫩茎无性系.
