UPLC－PDA法测定白花延龄草根茎及果实提取物中薯蓣皂苷元含量

摘要

With petroleum ether and H2 SO4 two-phase solvent hydrolysis ,Determination of diosgenin in the rhizome and fruit methanol extract of T .kamtschaticum Pall .using UPLC .chrom column:Waters BEH C18 (2.1mm × 100mm ,1.7μm );Detector:PDA .The mobile phase with a linear gradient elution of acetonitrile (A)-water(B):0min-8.0min ,50% -90% A;8.0min-8.1min ,90% ~100% A;8.1min-11.0min , 100% A;11.1min ,100% -50% A .Velocity :0.4mL/min;The column temperature :50℃ ;the detection wavelength:192nm;sample size:5μL . The results showed that ,the content of diosgenin rhizome methanol extracts was 0.03% ,and the fruit methanol extracts was 0.16% .%采用石油醚－硫酸溶液两相溶剂水解，UPLC－PDA法测定白花延龄草根茎和果实甲醇提取物中薯蓣皂苷元含量。色谱柱Waters BEH C18（2.1mm ×100mm ，1.7μm ）；检测器PDA。流动相乙腈（A ）－水（B ），线性梯度洗脱：0min～8.0min ，50％～90％A；8.0min～8.1min ，90％～100％ A；8.1min～11.0min ，100％ A；11.1min ，100％～50％ A。流速0.4mL／min；柱温50℃；检测波长192nm；进样量5μL。结果表明，白花延龄草根茎甲醇提取物中薯蓣皂苷元含量为0.03％，果实甲醇提取物中薯蓣皂苷元含量为0.16％。
