多型口蹄疫疫苗灭活时间和纯化的比较研究

             

摘要

[目的]为了探讨不同灭活时间及纯化对多型口蹄疫疫苗146 S、总蛋白量、抗原收获量及疫苗效力的影响.[方法]选取OM/Re-A/AF/Asia1 4种不同型的口蹄疫疫苗为研究对象,设定不同灭活时间(8、10、12、14和16 h),对每个时间点疫苗母液中146 S、总蛋白和抗原收获量进行检测和差异性分析;测量纯化前后146 S和总蛋白的变化趋势;测定不同灭活时间及纯化前后疫苗PD50数值进行判定效力.[结果]灭活不同时间点,146 S含量和抗原收获量随着时间先增高后平缓的趋势,146 S在14 h出现平缓,而抗原收获量在10h时开始平缓;146 S含量OM和AF型明显高于Re-A和Asia1型,抗原收获量Asia1型最高;总蛋白含量随着时间呈现先降低后平缓的趋势,AF型口蹄疫疫苗降低最明显;疫苗纯化前后相比分析发现,146 S含量明显升高,总蛋白含量睨显降低(P<0.01).4种疫苗PD50值随着纯化时间先增大后平稳,纯化后PD50显著提高(P<0.01),免疫效力增强.[结论]OM/Re-A/AF/Asia1 4种不同型的口蹄疫疫苗在纯化不同时间点,疫苗中146 S、抗原收获量和总蛋白含量呈规律性变化,且纯化前后146 S、总蛋白含量及PD50值差异性极显著,表明口蹄疫疫苗的灭活时间和纯化是影响疫苗效价的重要因素.%[Objective] The aim of this study is to investigate the different types of foot and mouth disease vaccine of 146 S,total protein contents,antigen harvest and vaccine efficacy by inactivate time and purify.[Method] OM/Re-A/AF/Asia1 four different types of foot and mouth disease vaccine were selected as the research objects,setting different inactivated time (8,10,12,14 and 16 hours),the difference of the 146 S,total protein contents and antigens harvest by each point-in-time were analyzed,and the changing trends of total protein and 146 S before and after purification were measured.The vaccine effect of PD50numerical at different time inactivated and before-after purification was identified.[Result] At the different time points of inactivated,146 S and antigen harvest with the time of increased first and then gentle trend,the 146 S appeared flat at 14 hours,but the antigen harvest began to gently at 10 hours;The 146 S content in OM and AF was significantly higher than that of Re-A and Asia1,and antigen harvest was the highest in Asia1;Purified over time and total protein content showed a trend of first decreases and then gentle,AF type of foot and mouth disease vaccine lowers was the most obvious;Compared of vaccine before and after purification,it was found that 146 S levels increased significantly,total protein content significantly decreased (P <0.01).Four types of vaccine PD5o value with the purification time firstly increases and then was smooth,purified PDs0 significantly increased (P <0.01),increasing the immune effect.[Conclusion] With the purification of OM/Re-A/AF/Asia1 four different types of foot and mouth disease vaccine at different time points,146 S,antigen harvest and total protein were regularly changed in vaccine,and the 146 S,total proteins and PD5o value difference significantly in before-after purification.This study indicated that the inactivated time and purification was one of the important factors in affecting vaccine titer.
