嗜蓝孢孔菌属(Fomitiporia)一新种

         

摘要

[目的]在山西雁北地区沙棘树腐木上采集到了野生嗜蓝孢孔菌属一新菌,研究其新菌的形态学特征、分子系统发育并确定新种.[方法]形态学观察采用肉眼观察标本以及制作Melzer、棉蓝和5%KOH 3种试剂的浮载切片,在显微镜下观察标本形态,分子系统发育分析采用将新种的ITS序列与其他已经报道的20种真菌ITS进行同源聚类分析,构建基于ITS的分子系统发育树.[结果]形态学特征为具有马蹄形担子果,盖状无柄,有时平伏反卷生长,单系菌丝系统;骨架菌丝在Melzer试剂和棉蓝试剂中呈负反应;担孢子近球形或球形,无色,厚壁等.分子系统发育分析结果表明新种与嗜蓝孢孔菌属亲缘关系较近,聚在一个分支中,并在系统发育树中形成了一个独立的亚分技.[结论]根据研究确认为嗜蓝孢孔菌属新种,定名为Fomitiporia yanbeiensisS.Guo&L.Zhou(登陆号GenBank:KT861405,Fungal name:FN570360).%[Objiective] A new Fomitiporia fungus,Fomitiporia sp.nov.,was collected from the Yanbei area Shanxi province China in the wild buckthorn trees rot wood.The purpose of this study was to identify the morphological characteristics,molecular phylogeny and new species of new strain.[Methods] Morphological observation was carried out by naked eye observation,and under the Nikon E80i microscope,3 kinds of reagents,Melzer,cotton blue and 5 % KOH,were used as slides to observe the microscopic shape,Molecular phylogenetic analysis was carried out by homologous clustering analysis of the new ITS sequences with 20 other reported fungal ITS,and a phylogenetic tree based on ITS was constructed.[Result] The morphological feature was that it has a horseshoe shaped cover basidiocarps,sessile,sometimes procumbent backscroll mycelium growth,single system;skeletal hyphae showed a negative reaction in the Melzer reagent and cotton blue reagent;basidiospore near spherical or spherical,colorless,thick wall etc.Molecular phylogenetic analysis showed that the new species had close relationship with the genus Cladosporium and clustered into one branch and formed an independent sub clade in the phylogenetic tree.[Conclusion] According to the study,it was confirmed as a new species of Fomitiporia genus and named Fomitiporia yanbeiensis S.Guo & L.Zhou (GenBank:KT861405,Fungal name:FN570360).The type specimen SXSYJBB1510 was stored in the Institute of edible fungi of Shanxi Academy of Agricultural Sciences,and the strain specimen CGMCC No.13380 has been deposited in China General Microbiological Culture Collection Center.
