骨髓基质干细胞移植对兔继发性肢体淋巴水肿的治疗效果观察

         

摘要

目的 观察骨髓基质干细胞(BMSCs)移植对兔继发性肢体淋巴水肿的治疗效果.方法 成功制作后肢淋巴水肿模型的新西兰大白兔20只,随机分为观察组(10只)及对照组(10只).观察组将BMSCs注射到兔右后肢手术创伤并射线照射区域水肿软组织(包括皮肤瘢痕及肌肉等组织),对照组同部位注射生理盐水,28 d后处死动物;观察两组肢体水肿体积(右后肢与左后肢体积的差值),水肿软组织微淋巴管计数及Flt-4 mRNA、VEGF-CmRNA及vWF mRNA表达变化.结果 与对照组比较,观察组肢体水肿体积减小,微淋巴管计数增加,且随时间变化而变化(P均<0.05);与对照组比较,观察组Flt-4 mRNA、VEGF-C mRNA表达升高(P均<0.01).结论 BMSCs移植可减小肢体水肿体积,促进新生淋巴管的生成及局部VEGF-C、Flt-4分泌.%Objective To observe the curative effect on secondary limb lymphedema of bone marrow stromal cells (BMSCs) transplantation in rabbits. Methods Operation and radiation were combined to gain 20 laboratory rabbit animal models, and then were divided into observation group( 10 animals) and control group( 10 animals) randomly . BMSCs were injected into edema tissue including scar tissue and muscle which were injured by surgery and radiation (edema tissue) of lymphedema limbs in observation group, and normal saline was injected in control group. The animals were sacrificed at 4 weeks after operation. The swelling of lymphedema limbs, micro-lymphatic vessel number and VEGF-C, Flt-4 and vWF mRNA in edema tissue were observed. Results Compared with the control group, the swelling of lymphedema limbs in the observation group reduced, and the micro-lymphatic vessel number increased (all P < 0. 05 ) ; Compared with the control group, the expression of Flt-4 mRNA and VEGF-C mRNA increased (all P <0.01). Conclusions BMSCs transplantation can promote secretion of VEGF-C and flt-4 in edema tissue, promote reconstruction of the lymphatic circulation reflux networks , ease and cure the limb lymphedema.
