猴头多糖对小鼠皮下移植肉瘤的抑制作用及机制探讨

         

摘要

Objective To observe the inhibitory effect of Hericium erinaceus polysaccharide on the mice subcutaneous transplanted sarcoma , and to explore its mechanism from apoptosis perspective .Methods S180 sarcoma cells were subcu-taneously injected into 50 mice to produce the models of subcutaneous tumor transplantation .After successful modeling , the tumor bearing mice were randomly divided into five groups , 10 in each group: the low-dose group , medium-dose group , high-dose group, model group and cyclophosphamide group .The low-dose, medium-dose and high-dose groups were given hedgehog polysaccharide 25, 50 and 100 mg/kg, orally, once a day, and for 10 days.The model group was given normal saline 0.2 mL/day orally and for 10 days.Cyclophosphamide was administered with 40mg/kg by intraperitoneal injection once a day for 5 days.All the mice were killed after 10 days of treatment .We calculated the inhibition rate of each group . The apoptosis in each group was detected by TUNEL assay .Caspase-3 protein was detected by immunohistochemical meth-od.Results The tumor inhibition rates in the low-dose group, medium-dose group, high-dose group and cyclophospha-mide group were 36.36%, 57.57%, 43.93%and 74.24%.The tumor inhibition rate in the cyclophosphamide group was higher than those in the other groups , and in the medium-dose group , the tumor inhibition rate was higher than those of the low-dose group and high-dose group (all P <0.05).The number of apoptotic cells and Caspase-3 protein expression were as follows:cyclophosphamide group >medium-dose group >high-dose group >low-dose group >model group ( all P<0.05).Conclusion Hericium erinaceus polysaccharide can inhibit the growth of xenograft sarcoma , and the dose of 50 mg/kg dose is the best .This mechanism may be related to its up-regulation of Caspase-3 protein expression and thereby promoting tumor cell apoptosis .%目的:观察猴头多糖对小鼠皮下移植肉瘤的抑制作用,并从细胞凋亡角度探讨其作用机制。方法将50只小鼠腋下皮下注射S180肉瘤细胞,制作肿瘤移植模型。造模成功后随机分为低剂量组、中剂量组、高剂量组、模型组、环磷酰胺组各10只,低剂量组、中剂量组、高剂量组分别予猴头多糖25、50、100 mg/kg灌胃,1次/d,共干预10 d;模型组予生理盐水0.2 mL/d灌胃,连续10 d;环磷酰胺组予环磷酰胺40 mg/kg腹腔注射,1次/d,共干预5 d。于第10天处死各组小鼠,计算抑瘤率,TUNEL法检测凋亡细胞,免疫组化法检测Caspase-3蛋白。结果低剂量组、中剂量组、高剂量组、环磷酰胺组抑瘤率分别为36.36%、57.57%、43.93%、74.24%,环磷酰胺组抑瘤率明显高于其余各组,中剂量组抑瘤率高于低剂量组、高剂量组,P均<0.05;凋亡细胞数及Caspase-3蛋白表达为环磷酰胺组>中剂量组>高剂量组>低剂量组>模型组,P均<0.05。结论猴头多糖可抑制小鼠皮下移植肉瘤的生长,以50 mg/kg剂量作用最佳;该作用与其增加Caspase-3蛋白表达、促进肿瘤细胞凋亡有关。
