胰腺导管细胞癌组织CSF3R基因表达对患者预后的影响及其机制

     

摘要

目的 探讨粒细胞集落刺激因子3受体( CSF3R)基因表达对胰腺导管细胞癌患者预后的影响及其机制.方法 从GEO数据库中下载包含CSF3R表达数据的人类胰腺导管细胞癌表达谱,包括132例份胰腺导管细胞癌样本.通过R2分析平台,以Kaplan-Meier法绘制生存曲线,分析CSF3R表达与患者预后的关系,筛选出与CSF3R表达显著相关的基因并进行KEGG通路分析与GO分析.结果 CSF3R表达与患者总体生存率呈负相关(P<0.05),CSF3R高表达患者的总体生存率低于低表达患者(P<0.05).检测出与CSF3R表达呈显著相关的基因共6 521个,其中正相关基因3 040 个、负相关基因3 481 个. KEGG分析发现,有27 个高表达基因( C1QA、C1QB、C1QC、 C2、 C3AR1、 C5、 C5AR1、 FCAR、 FCGR2A、 FCGR2B、 FCGR3A、 FCGR3B、 FGG、 FPR1、 FPR2、 FPR3、HLA-DMB、HLA-DQA1、HLA-DQB1、ICAM1、ITGAM、ITGB2、MASP1、MASP2、MBL2、PLG、SELPLG)仅涉及1个信号通路,即金黄色葡萄球菌感染. GO分析发现,CSF3R及其相关基因主要与细胞对刺激的反应、免疫反应、RAS调控、胰腺发育等相关.结论 CSF3R表达与胰腺导管细胞癌患者总体生存率相关,其可能是预测患者预后的潜在分子标记物.%Objective To investigate the relationship between the expression of granulocyte colony-stimulating factor-3 receptor (CSF3R) gene and the prognosis of patients with pancreatic ductal adenocarcinoma , and to analyze the correlated genes and cellular pathways.Methods The corresponding cancer datasets included 132 samples were extracted from the public database GEO to obtain the patients’ information and mRNA expression data.The survival curve was drawn by Kap-lan-Meier method using the R2 bioinformatics analysis platform, and the relationship between the expression of CSF 3R and the prognosis of the patients was analyzed.Genes significantly associated with CSF 3R expression were enriched and analyzed for KEGG pathways and GO pathways.Results The datasets showed a significant negative correlation between CSF 3R mRNA expression level and the overall survival rate (P<0.05).The overall survival rate was higher in patients with high CSF 3R expression than in patients with low CSF3R expression (P<0.05).In addition, there were 6 521 genes associated with CSF3R expression, during which, 3 040 genes were positively correlated and 3 481 genes were negatively correlated.KEGG analysis showed that 27 gene were highly expressed (C1QA, C1QB, C1QC, C2, C3AR1, C5, C5AR1, FCAR, FCGR2A, FCGR2B, FCGR3A, FCGR3B, FGG, FPR1, FPR2, FPR3, HLA-DMB, HLA-DQA1, HLA-DQB1, ICAM1, ITGAM, ITGB2, MASP1, MASP2, MBL2, PLG, SELPLG), and they involved only one signaling pathway, staphylococcus aureus infection.GO analysis found that CSF3R and its related genes were mainly related to cell response to stimulation, immune response, RAS regu-lation, and pancreas development.Conclusion CSF3R expression is negatively correlated with the overall survival rate of pa-tients with pancreatic ductal adenocarcinoma, and it may be a potential molecular marker for the study of prognosis of patients.
