半胱氨酸保护基S-Phenylacetamidomethyl(Phacm)的合成及应用

摘要

Disulfide bonds formed by cysteine are an important structural element in many proteins and peptides. Sulfydryl group of cysteine needs to be protected in advance by relative protecting group during formation of the disulfide bonds, otherwise the nucleophilic thiol will occur various side reactions. In this paper, the synthesis, removal, compatibility and application of a new cysteine protective group S-Phacm are introduced. The research prospect is expected.%由半胱氨酸形成的二硫键在许多蛋白质和多肽中是一类非常重要的结构元素.合成此类二硫键时半胱氨酸的巯基必须要预先被保护基保护起来,否则具有亲核性的巯基会发生许多副反应.本文介绍了一种新的半胱氨酸保护基S-Phacm的合成、脱除、兼容性及应用,并对其研究前景进行了展望.
