γ射线联合2450MHz电磁辐射对C3H10T1/2细胞生长和恶性转化的影响

         

摘要

Objective :To investigate the interactive effects of 2450 M Hz microwave and γ‐rays radiation on C3H10T1/2 cells growth ,malignant transformation ,Cell cycle and apoptosis .Methods:C3H10T1/2 cells were ir‐radiated with 0 ,15 ,30 ,50mW/m2 2450 M Hz microwave and 6Gy γ‐ray radiation .this study was to investigate the possible effects of 2450 MHz microwave at different doses on γ‐ray induced transformation in C3HlOT1/2 cells ;Cell cycle and apoptosis was assessed by flow cytometry .Bax expression of cells were detected by immuocytochemical staining with image analysis .Results :Compared with control group ,microwave radiation group of cell survival and cell transformation was not changed .there was a significant increase in malignant transformation at γ‐ray plus mi‐crowave radiation group .cells S stages were significantly decreased ,while cells in G0/G1 stages were significantly in‐creased at 3 h .Moreover ,the amount of apoptosis cells in combined radiation group was more than that in γ‐ray irra‐diation group .In the early days after irradiation ,Bax protein expression was raised .Conclusion :γ‐ray combined with microwave could induce malignant transformation and apoptosis of C3H10T1/2 cells ,and microwave could enhance the effect of γ‐ray .The expressions of Bax in harmony may play a role in apoptosis of C3H10T1/2 cell .%目的:观察γ射线和2450 M Hz电磁联合辐射对C3H10T1/2小鼠胚芽细胞增殖能力、恶性转化、细胞周期和细胞凋亡的影响。方法:将C3H10T1/2细胞按电磁辐射强度随机分为对照组(0mW/m2)、电磁辐射低剂量组(15mW/m2)、中剂量组(30mW/m2)和高剂量组(50mW/m2) ,联合组同时给予6Gy的γ射线照射。检测C3H10T1/2小鼠胚芽细胞转化、细胞周期和凋亡情况;用免疫组化和图像分析技术检测细胞Bax蛋白表达。结果:与对照组相比,单纯电磁辐射处理的细胞存活率和细胞转化未发生改变,γ射线与高剂量电磁辐射联合组存在恶性转化。γ射线组和复合组于辐射后3h S期细胞显著减少,G0/G1期细胞比例明显升高,电磁辐射组细胞凋亡率轻度增高,而γ射线组和复合组则升高较为显著,且复合组引起细胞的凋亡率升高较单纯γ射线组更明显,照射后Bax表达增强。结论:γ射线与电磁辐射复合照射后C3H10T1/2细胞发生恶性转化、细胞周期和凋亡,2450M Hz电磁辐射可进一步加强γ射线对细胞恶性转化、细胞周期及凋亡的诱导作用。Bax在γ射线和2450M HZ电磁联合辐射导致C3H10T1/2细胞发生凋亡且较单纯组加重方面起重要作用。
