以西洋参种子为材料,研究了6-BA或NAA与GA3组合及培养基中赤霉素灭菌方式对不同后熟程度种胚无菌苗培养的影响,筛选无菌苗培养的最佳培养基.结果表明:(1)在培养基中,同一GA3浓度下,添加NAA比6-BA更能促进种胚萌发,且形成的无菌苗子叶不衰老;(2)在同一培养基中,后熟程度高的种胚比后熟程度低的种胚萌发率高,长势好;(3)赤霉素应采取过滤灭菌方式加入培养基;(3)无菌苗培养最佳培养基为MS+2 mg/L NAA+5 mg/L GA3.%In this paper,American ginseng seeds were used to study the effects of GA3 combined with 6-BA or NAA and GA3 sterilization methods on the embryos with different after-ripening into seedlings, and the best medium for aseptic seedlings culture were selected. The results showed that: (1) when NAA was added, there was higher germination rate on medium containing the same GA3 concentration, and the cotyledons of aseptic seedlings was not aging. ( 2 ) the embryos with higher degree of after-ripening had better germination and growth than the lower degree after-ripening embryos in the same medium. ( 3 ) the better medium for aseptic seedlings culture was MS +2 mg/L NAA +5 mg/L GA3.
