This paper discussed improvement of purity detection of hybrid rice seeds by SSR molecular markers from multiple aspects,such as optimization of the DNA extraction method,application of the multiple spotting electrophoresis technique and determination of the proper sample size,which may make the indoor purity detection more reliable and higher efficient.The results showed that it is feasi-ble to extract DNA of brown rice by TPS method and analyze DNA with third spotting in electropho-resis.Also,the sample size of 192-288 is suitable for detection of indoor seed purity%通过改进DNA提取方法,利用多层点样的电泳技术和样本量确定等方面,探讨 SSR分子标记检测杂交稻种纯度技术的改良,以期能提高室内纯度检测的可靠性及效率.结果表明:利用TPS法提取糙米DNA和3层点样的电泳技术是可行的;192~288的样本量较适合室内种子纯度检测.
