山药中降糖活性成分的筛选与比较

     

摘要

Objective: To study the effect of extracts of dioscorea rhizome-polysaccharides, saponins and flavones to HepG2 cell with the subject of consumption glucose affection and to hypoglycemic action effect to the diabetic mice model established intra-abdominal injection of alloxan in order to compare the three components' hypoglycemic bioactive.Methods : using the D101 macroporous resin adsorption process for the ethanol extracts of dioscoreae rhizome,elution with different concentration of ethanol.Depurating get 50% flavones content or so, and 60% saponins,and get polysaccharides from extracts of dioscoreae rhizome by water.Then use HepG2 cell and the diabetic mice as model to study three components hypoglycemic affections.Results: It demonstrated that saponins could lead to adding consumption of glucose in unit cell and make model dose-dependent.Dioscoreae rhizome,saponins and flavones could decrease the water drinking volume of diabetic mice and level of glucose.Dioscoreae rhizome was best.It concluded that three components showed synergistic effect among dioscoreae rhizome' s dhypoglycemic bioactive base on the experiment.%目的:研究山药中多糖、皂苷、黄酮粗提物对HepG2细胞的葡萄糖的消耗作用,对四氧嘧啶诱导的糖尿病小鼠的降糖作用,以此来综合比较三种成分的降糖效果.方法:山药乙醇提取物经(D101)大孔树脂柱吸附乙醇梯度洗脱、分离纯化得到50%含量的总黄酮和60%含量的总皂苷,山药水提物纯化得到多糖,然后分别以HepG2细胞、糖尿病小鼠为模型研究了三种成分的降血糖效果.结论:皂苷可增加单位细胞的葡萄糖消耗量,呈现剂量依赖性.山药原粉、皂苷和黄酮能显著降低糖尿病小鼠的饮水量和血糖水平,其中原粉最明显.推测在山药的降血糖作用中三种成分存在协同效应.
