依达拉奉对过氧化氢致血管内皮细胞氧化损伤的保护作用

         

摘要

Objective To investigate the protective effect of edaravone on endothelial cell injuried by hydrogen peroxide (H2O2). Methods Human umbilical vein endothelial cell (HUVEC) were cultured in vitro and were injuried by H2O2 as oxidative stres model,so as to observe the protective effects of edaravone by means of determining the indexes of oxidative stress which included the contents of malonyldialdehyde(MDA),the activity of superoxide dismutase(SOD) and total antioxygen capability(T-AOC). Results The content of MDA was markedly decreased,the activity of SOD and the T-AOC was significantly increased in edaravone groups compared with H2O2 group. And all these effects of edaravone groups were dose-dependent. Conclusion Edaravone has protective effect on H2O2 injuried HUVEC,and the protective effect of edaravone may be due to its reducing the degree of oxidative damage and improving the antioxidant ability on HUVEC.%目的:探讨依达拉奉对过氧化氢(H2O2)致内皮细胞氧化损伤的影响。方法体外培养人脐静脉内皮细胞(HUVEC),采用H2O2作为外源性自由基生成系统,模拟血管内皮细胞氧化应激损伤模型,观察不同剂量依达拉奉对H2O2所致内皮细胞的影响。通过检测各组丙二醛(MDA)的含量反应细胞的损伤程度,检测超氧化物歧化酶(SOD)的活性及总抗氧化能力(T-AOC)反应细胞的抗氧化能力。结果依达拉奉各浓度组均明显降低MDA的含量,提高SOD的活性及总T-AOC,上述作用随药物浓度增加呈增强趋势。结论依达拉奉能降低内皮细胞的氧化损伤程度,提高内皮细胞抗氧化能力。
