HPLC法同时测定不同产地通关藤中通关藤苷GIH的含量

         

摘要

目的 采用HPLC法同时测定不同产地通关藤中通关藤苷G、I、H的含量.方法 色谱柱为资生堂MG-G18(3.0 mm×100 mm,3.0 μm),流动相为乙腈-0.1%的甲酸水溶液(40∶60,V/V),流速:0.5 ml·min-1,波长:230 nm,柱温:25℃,进样量:5μl,外标法定量.结果 3种通关藤苷在25 min内基线分离,方法学验证表明,线性关系良好(r>0.9999),日内日间精密度均小于2.0%,回收率在98%~102%,稳定性和重复性RSD也均小于2.0%,符合方法学要求.结论 本方法测定了云南、贵州、广西、四川4个产地中3种通关藤苷的含量,快速简便,可为不同产地通关藤药材的质量控制提供参考依据,也为后期开展药材中通关藤苷类有效成分的体内代谢及药动学研究奠定了良好的基础.%Objective To establish an HPLC method for simultaneous determination of three types of tenacissosides in Marsdenia tenacissima from different species.Methods The HPLC conditions were as follows:SHISEIDO MG-C18 column (3.0 mm× 100 mm,3.0 μm),the mobile phase was acetonitrile and water containing 0.1% formic acid(40:60,V/V),flow rate was 0.5 ml ·min-1,UV detection wavelength was 230 nm,column temperature was 25 ℃,injection volume was 5 μl,and the external standard method was used for quantitative analysis.Results Three types of tenacissosides were separated at baseline within 25 min.The standard curves had good linearity,the RSD of intraday and interday precision was less than 2.0%,the recovery was between 98% and 102%,and the RSD of stability and repeatability was also less than 2.0 %.The results showed that this method could meet the requirements of method validation.Conclusion Three types of tenacissosides in Marsdenia ten acissima from 4 different species (Yunnan,Guizhou,Guangxi,Sichuan) were determined.This method is quick and simple,which can be used for quality control of Marsdenia tenacissima,thus facilitating the metabolism and PK study of tenacissosides in the future.
