Objective: To investigate the effect of tanshinone ⅡA (Tan ⅡA)on the vasculogenic mimicry in breast cancer cell line MDA-MB-231 in vitro.Methods: Cultured cells were divided into Tan ⅡA group and control group.The effect of proliferation and tube formation in MDA-MB-231 cells were detected by MTT assay and tube formation assay respectively in vitro.The mRNA and protein levels of various genes which related to vasculogenic mimicry formation were detected by RT-PCR and Western-blot.Results: Tube formation could be inhibited by Tan ⅡA.The mRNA levels and the protein levels of VEGF were down-regulated while the concentration of Tan ⅡA was increasing.Conclusion: Tan ⅡA can inhibit the vasculogenic mimicry formation of MDA-MB-231 in vitro by downregulating VEGF protein expression.%目的:研究丹参酮ⅡA对乳腺癌细胞株MDA-MB-231体外血管生成拟态的影响.方法:将培养的MDA-MB-231细胞分为丹参酮ⅡA组及对照组,应用MTT法检测细胞增殖活力的变化,体外管状形成试验检测管道形成能力的变化,通过RT-PCR和Western-blot技术检测细胞内与血管拟态形成相关基因表达的变化.结果:丹参酮ⅡA对MDA-MB-231细胞增殖有明显抑制作用,并抑制体外管道形成和肿瘤细胞VEGF的mRNA表达.结论:丹参酮ⅡA能抑制MDA-MB-231细胞的体外血管生成拟态的形成和细胞增殖,其机制可能是通过抑制VEGF的表达而实现.
