京尼平苷对LPS诱导的星形胶质细胞活化及炎性反应的抑制作用

         

摘要

Objective To determine the inhibitory effect of geniposide on the activation and inflammatory response of lipopolysaccha-ride(LPS)induced astrocytes.Methods The LPS induced primary cultured astrocytes as cell model were randomly divided into control group,LPS group,LPS+ resveratrol group and LPS+ different concentrations of geniposide group.GFAP markers and NF-level p65 kappa B(NF-κB)activation of astrocytes were detected by Western Blot.The inflammatory cytokines were detected by ELISA.Results Geniposide could inhibit astrocyte activation induced by LPS.Compared with LPS group,IL-1β,IL-6 and TNF-αwere decreased by geniposide ,as well as protein level of NF-κB p65 .Conclusion LPS induced astrocyte activation and proinflam-matory cytokines expression could be inhibited by geniposide,this effect may act by inhibiting NF-κB activation.%目的 明确京尼平苷对脂多糖(LPS)诱导的星形胶质细胞活化及炎性反应的抑制作用.方法 以LPS诱导原代培养的星形胶质细胞为细胞模型,随机分为对照组、LPS组、LPS+白藜芦醇组和LPS+不同浓度京尼平苷组,采用Western Blot法检测星形胶质细胞活化标记物GFAP 水平及NF-κB p65水平,ELISA法检测致炎细胞因子水平.结果 京尼平苷可显著抑制LPS诱导的星形胶质细胞活化,降低TNF-α、IL-1β和IL-6的表达,同时NF-κB p65蛋白的表达也明显降低.结论 京尼平苷能通过NF-κB途径抑制LPS诱导的星形胶质细胞活化,并抑制致炎细胞因子水平.
