脾阴虚痴呆大鼠不同脑区Snk-SPAR路径及滋补脾阴方药调控作用研究

     

摘要

目的:观察脾阴虚痴呆大鼠不同脑区树突棘相关的Rap -特异性GTPase -活化蛋白和血清诱导激酶mRNA表达的变化及滋补脾阴方药对其调控作用.方法:采用逆转录聚合酶链反应检测各脑区SPAR、Snk mRNA表达变化.结果:痴呆组和脾阴虚痴呆组大鼠各脑区SPAR mRNA表达明显减弱(P<0.05),Snk mRNA表达呈上调趋势;脾阴虚痴呆+ZBPYR组大鼠各脑区SPAR mRNA表达明显增强(P<0.05),Snk mRNA表达呈下调趋势.结论:ZBPYR能使脾阴虚痴呆组大鼠各脑区SPARmRNA表达增强,Snk mRNA表达下调,提示ZBPYR具有保护并维持突触和树突棘正常形态的作用,这一作用机制可能与Snk-SPAR信号途径传递有关.%The study is aimed to observe the effect of Zibu Piyin Recipe (ZBPYR) on the expression of SPAR and Snk mRNA in different brain regions of spleen-yin deficiency Alzheimer's disease model rats. Levels of SPAR and Snk mRNA expression were detected by using RT-PCR method. The result showed that level of SPAR mRNA ex pression in different brain regions in AD group and spleen-yin deficiency AD group decreased significantly (P< 0.05). And the level of Snk mRNA expression was up-regulated. The level of SPAR mRNA expression in different brain regions in ZBPYR treatment group increased significantly (P<0.05), while the level of Snk mRNA expression was down-regulated. It is concluded that the level of SPAR mRNA expression increased significantly while the level of Snk mRNA expression decreased after treated with ZBPYR. It indicates that ZBPYR has the function of protecting and maintaining morphology and structure of synapses and dendritic spines. The mechanism can be associated with its actions in the inhibition of the Snk-SPAR signal pathway.
