益气健脾方对慢性阻塞性肺疾病大鼠肺功能及血清TNF-α和IL-8的影响

         

摘要

目的 观察益气健脾方对慢性阻塞性肺疾病大鼠肺功能及血清中肿瘤坏死因子-α (TNF-α) 、白细胞介素-8 (IL-8) 水平的影响.方法按随机数字表法, 将60只健康雄性SD大鼠分为正常组、模型组、地塞米松组、中药低剂量组、中药中剂量组、中药高剂量组, 每组10只.除正常组外, 其余组均采用烟熏加气管内注射脂多糖的方法进行慢性阻塞性肺疾病大鼠造模.在第2次注射脂多糖后, 地塞米松组给予地塞米松灌胃, 中药低、中、高剂量组分别给予相应剂量益气健脾方浓缩液灌胃, 模型组给予等剂量生理盐水灌胃, 每天1次, 至烟熏结束.实验结束后, 采用小动物肺功能测定仪测定各组大鼠肺功能, 用酶联免疫吸附法检测各组大鼠血清中TNF-α和IL-8水平.结果 各造模组大鼠FEV0. 3/FVC均明显低于空白组 (P均<0. 05) , 地塞米松组和中药各剂量组FEV0. 3/FVC均明显高于模型组但仍低于空白组 (P均<0. 05) , 地塞米松组及中药各剂量组比较差异均无统计学意义 (P均> 0. 05) .各造模组大鼠血清IL-8、TNF-α水平均明显高于空白组 (P均<0. 05) ;地塞米松组、中药各剂量组大鼠血清IL-8、TNF-α水平均明显低于模型组 (P均<0. 05) ;地塞米松组及中药各剂量组大鼠血清IL-8水平比较差异均无统计学意义 (P均> 0. 05) ;地塞米松组、中药低剂量组大鼠血清TNF-α水平比较差异无统计学意义 (P> 0. 05) ;中药中剂量组、中药高剂量组大鼠血清TNF-α水平均明显低于地塞米松组 (P均<0. 05) ;中药各剂量组间大鼠血清TNF-α水平比较差异均无统计学意义 (P均> 0. 05) .结论 益气健脾方能够降低慢性阻塞性肺疾病大鼠血清中TNF-α和IL-8水平, 减轻气道炎症, 改善肺功能.%Objective It is to observe the effects of Yiqi Jianpi Decoction on lung function and serum levels of tumor necrosis factor-α (TNF-α) and interleukin-8 (IL-8) in rats with chronic obstructive pulmonary disease. Methods According to the random number table method, 60 healthy male Sprague-Dawley rats were divided into normal group, model group, dexamethasone group, low-dose Chinese medicine group, medium-dose Chinese medicine group and high-dose Chinese medicine group, with 10 rats in each group. Except the normal group, the rats in the other groups were given lipopolysaccharide by tube injection plus smoking to establish the models of chronic obstructive pulmonary disease. After the second injection of lipopolysaccharide, the dexamethasone group was given dexamethasone by lavage, and the low, medium and high doses of traditional Chinese medicine groups were given the corresponding doses of concentrated solution of Yiqi Jianpi Decoction by lavage, the model group was given the same dose of saline by lavage, once a day, until the end of the smoke. After the experiment, the lung function of each group was measured by small animal lung function tester. The levels of TNF-α and IL-8 in the serum of each group were detected by enzyme-linked immunosorbent assay. Results The FEV0. 3/FVC of each pathologic group was significantly lower than that of the blank group (P < 0. 05). The FEV0. 3/FVC of the dexamethasone group and the traditional Chinese medicine groups were significantly higher than that of the model group, but still lower than the model group. There was no significant difference between the dexamethasone group and the traditional Chinese medicine groups (P> 0. 05). The serum levels of IL-8 and TNF-α in the rats of each pathologic group were significantly higher than those in the blank group (P < 0. 05). The levels of serum IL-8 and TNF-α in the dexamethasone group and the traditional Chinese medicine groups were significantly lower than those in the model group (P < 0. 05). There was no significant difference in serum IL-8 level between dexamethasone group and the traditional Chinese medicine groups (P>0. 05). There was no significant difference in serum TNF-α level between the low dose Chinese medicine group and dexamethasone group (P> 0. 05) , but the levels of serum TNF-α in middle and high dose of Chinese medicine groups were significantly lower than that in the dexamethasone group (P < 0. 05). There were no significant differences in serum TNF-αlevel between very dose of traditional Chinese medicine group (P> 0. 05). Conclusion Yiqi Jianpi Decoction can reduce the levels of TNF-α and IL-8 in serum, reduce airway inflammation and improve lung function in the rats with chronic obstructive pulmonary disease.
