姜黄素对血管紧张素Ⅱ诱导新生大鼠心肌细胞肥大的保护作用

         

摘要

Objective It is to study the effect of curcumin ( Cur ) on the hypertrophy of cardiomyocytes in neonatal rat induced hy angiotensin Ⅱ ( AngⅡ ) and to approach the possihle action mechanism. Methods The cardiomyocyte hypertrophy cell model was induced with AngⅡ stimulating. The contigency treatment was given with Cur 2. 5 × 10 -5 mol/L. The cell size was measured by phase contrast microscope and the total protein content of cardiomyocyte was detected, which was regarded as the indexes of hypertrophy of cardiomyocytes. The cardiomyocytes was incubated with fura - 3/AM. The Ca2 + concentration of intra-cellular (( Ca2+ )i ) was determined and analyzed with fluorescence microscope and Felix software. The protein expression of calcineurin ( CaN ) was detected with Western blot. Results In AngⅡ group, the cell diameter was aggrandized and the total protein content was increased, which was significant difference compared with control group ( P < 0. 01 ). Cur 2. 5 × 10 -5 mol/L could lower the hypertrophy degree of cardiomyocytes ( P <0. 01 ). the ( Ca2+ )i ( P <0. 01 ) and the protein expression of CaN ( P < 0. 05 ). Conclusion Cur can inhibit the hypertrophy of cardiomyocytes in neonatal rat induced by AngⅡ. The mechanism maybe relate to the inhibition of Ca2 + /CaN signaling transduction pathway.%目的 研究姜黄素(Cur)对血管紧张素Ⅱ(AngⅡ)诱导新生大鼠心肌细胞肥大的保护作用并探讨其可能作用机制.方法 AngⅡ刺激新生大鼠心肌细胞,制备心肌细胞肥大模型,用Cur 2.5×10-5mol/L进行预防性治疗.采用相差显微镜测量细胞大小及测定心肌细胞总蛋白含量作为心肌细胞肥大的指标;fura-3/AM孵育心肌细胞,利用荧光显微镜及Felix软件分析测定细胞内钙离子浓度((Ca2+)i);Western blot检测钙调神经磷酸酶(CaN) 蛋白表达.结果 AngⅡ组细胞直径增大,总蛋白含量增加,与对照组相比有显著性差异(P均<0 01);Cur 2.5×10-5mol/L能够降低心肌细胞肥大程度(P<0.01)、(Ca2+)i(P<0.01)及CaN蛋白表达(P<0.05).结论 Cur对AngⅡ诱导心肌细胞肥大有明显的保护作用, 其机制可能与抑制Ca2+/CaN信号转导通路有关.
