补阳还五汤联合神经干细胞移植对脊髓损伤大鼠后肢功能恢复的影响

         

摘要

Objective It is to observe the combination of Buyang-Huanwu decoction and transplanted neural stem cells on the functional recovery of the hind limb locomotion in spinal cord transected rats. Methods After rat models were successfully established by T10 spinal cord segment transected completely, they were randomly divided into control group, Chinese medicine group, neural stem cells ( NSCs ) group, Chinese medicine plus NSCs transplanted group. Eight weeks after operation, BBB score and electrophysiology tests were performed, and the changes of tissue structure of spinal cord. Results The hind limbs of rats were completely paralysis in control group. Locomotion of hind limbs was observed in other groups of rats. BBB score of Buyang-Huanwu decoction plus NSCs transplanted group was higher than that of other groups. The hind limbs of rats were failed to climb on grids in control group. Five rats climbed to the platform through incline grids in NSCs transplanted group, four rats in Buyang-Huanwu decoction group and seven rats in Buyang-Huanwu decoction plus NSCs transplanted group. The latency prolonged and the amplitude became small in the cortical motor evoked potential ( CMEP ) after spinal cord injure in control group. The latency and the amplitude of Buyang-Huanwu decoction plus NSCs transplanted group were improved obviously compared with that of NSCs transplanted group and Buyang-Huanwu decoction group. Conclusion Combination of Buyang-Huanwu decoction and transplanted NSCs may promote the functional recovery of the hind limb locomotion in rat spinal cord transected completely.%目的 观察补阳还五汤与神经干细胞移植联合应用对脊髓损伤大鼠后肢运动功能恢复的影响.方法 大鼠胸10 脊髓段全横断损伤造模成功后,随机分为对照组、中药组、干细胞组、中药干细胞组.各组在第8周时进行行为学和电生理检测并观察脊髓组织结构变化情况.结果 对照组大鼠后肢完全瘫痪,其余各组均能观察到大鼠后肢的运动,中药干细胞组BBB 分数高于其他组;对照组大鼠后肢不能爬行网格,干细胞组5只、中药组4只和中药干细胞组7只大鼠能通过爬行网格攀上平台;脊髓损伤后,对照组的皮质运动诱发电位的潜伏期增长和峰值变小,中药干细胞组的潜伏期和峰值均较中药组和干细胞组有明显的改善.结论 补阳还五汤与神经干细胞移植联合应用能促进脊髓损伤大鼠后肢运动功能的恢复.
