高压氧对神经病理性疼痛大鼠疼痛行为学的影响

Objective It is to observe the effects of hyperbaric oxygen ( HBO) on pain-related behaviours in the rats with neuropathic pain .Methods Thirty-two male SD rats were randomly divided into four groups ( eight rats per group ) , inclu-ding:a sham operation group (group S), in which the sciatic nerve was exposed but not ligated and HBO was not performed ;a sciatic nerve with CCI group (CCI group), in which the sciatic nerve was ligated but HBO was not performed;a HBO pre-treatment group ( pre-HBO group) , in which HBO was initiated 12 h before CCI;and a HBO post-treatment group ( post-HBO group), in which HBO was initiated 12 h after CCI.Baseline mechanical withdrawal thresholds (MWT) and thermal withdrawal latencies ( TWL) were measured in all rats one day before CCI .Pain-related behaviours were observed in one , two, three, seven, 14, 21 and 28 days after CCI,MWT and TWL were measured .Results The lame, licking foot of neuralgia symptoms appeared in the rats after CCI .There was no significant difference in MWT and TWL among the groups before CCI . MWT and TWL were significantly lower in the pre -HBO, post-HBO and CCI groups compared with group S at each time point (P<0.05), and the reduction of MWT and TWL were highest in the CCI group .Compared with the CCI group, the me-chanical withdrawal threshold was significantly increased and thermal withdrawal latency was significantly extended in the pre-HBO and post-HBO groups (P<0.05).Conclusion HBO has preventive and therapeutic effects on neuropathic pain and HBO pretreatment was more effective than HBO post -treatment .%目的:观察高压氧对神经病理性疼痛大鼠疼痛行为学的影响。方法将32只成年雄性SD大鼠随机分成4组,每组8只。假手术组暴露坐骨神经但不结扎,不进行高压氧治疗;坐骨神经结扎组结扎坐骨神经,但不进行高压氧治疗;高压氧预处理组于坐骨神经结扎术前12 h行高压氧治疗;高压氧后处理组于坐骨神经结扎术后12 h行高压氧治疗。术前1 d测量各组大鼠机械痛阈值(MWT)和热痛阈值(TWL),术后第1,2,3,7,14,21和28天观察各组大鼠疼痛反应行为并测量MWT和TWL。结果坐骨神经结扎术后大鼠出现跛足、舔脚等神经痛症状。坐骨神经结扎术前各组MWT和TWL值比较差异无统计学意义。高压氧预处理组、高压氧后处理组和坐骨神经结扎组各时间点MWT及TWL值均明显低于假手术组(P均<0.05),且坐骨神经结扎组下降幅度最大。高压氧预处理组和高压氧后处理组各时间点MWT及TWL值均明显高于坐骨神经结扎组( P均<0.05)。结论高压氧对神经痛的发生有预防和治疗作用,且高压氧预处理比高压氧后处理更有效。
