买朱尼对骨性关节炎大鼠关节软骨中 Wnt/β-catenin信号通路的调控作用研究

         

摘要

Objective It is to study the regulatory function of Uygur medicine Maizhuni on Wnt /β-catenin pathway in rat osteoarthritis models .Methods Knee osteoarthritis model was constructed using modified Hulth method .30 healthy SD rats were randomly divided into sham group , model group and Maizhuni group with 10 rats in each group .From the second week , rats were administered with Uygur medicine Maizhuni (10.31 mg/(kg· d)) or saline by gavage for 4 weeks.General scores and Mankin scores of rat knee joint cartilage in the three groups were evaluated .Quantitative real -time PCR and Western blotting were used to determine the expression of β-catenin, GSK-3βand Wnt-3a in the Wnt/β-catenin pathway .Re-sults Compared to the sham group , the general scores and Mankin scores of rat knee joint cartilage degeneration in Maizhuni group was significantly increased .Compared to the model group , the degree of rat knee joint cartilage degeneration in Maizhuni group was obviously alleviated .The cartilage general score and Mankin score in Maizhuni group was lower than that in the model group (P<0.05).On the molecular lever, we found that Maizhuni treatment significantly reduced the expression of β-catenin, GSK-3βand Wnt-3 in the knee joint chondrocytes (P<0.05).Conclusion Uygur medicine Maizhuni could protect the articular cartilage by regulation of Wnt/β-catenin pathway .%目的:观察维药买朱尼对骨性关节炎模型大鼠关节软骨中Wnt/β-catenin信号通路的调控作用,探讨买朱尼对软骨细胞的保护作用机制。方法将30只SD大鼠随机分为假手术组、模型组和买朱尼组,每组10只。模型组和买朱尼组采用改良Hulth造模法建立膝骨关节炎模型,建模第2周开始买朱尼组灌胃买朱尼10.31 mg/(kg· d),模型组和假手术组灌胃等量生理盐水,均连续4周。末次灌胃后24 h取3组大鼠膝关节软骨标本进行大体评分和组织学评分,采用实时荧光定量PCR和免疫印迹法检测Wnt/β-catenin通路中β-cate-nin、GSK-3β和Wnt-3a的表达水平。结果模型组大鼠膝关节软骨标本大体评分、软骨组织学Mankin评分及软骨细胞中β-catenin、GSK-3β、Wnt-3a蛋白及mRNA表达水平均明显高于假手术组(P均<0.05),而买朱尼组均明显低于模型组(P均<0.05)。结论维药买朱尼可以减轻大鼠膝关节软骨退变的程度,其可能通过调节Wnt/β-catenin信号通路来发挥保护关节软骨细胞的作用。
