HPLC法比较不同产地白花蛇舌草中两种香豆素类成分的含量

         

摘要

Objective: To compare the contents of scopoletin and hedyotiscone B in Hedyotis diffusa from different areas. To provide reference for the perfection of quality standards for H. diffusa.Method: The separation was accomplished on an Agilent Eclipse SB-C18 column (4.6 × 250 mm, 5 μm). The mobile phase was MeOH- 0.05%formic acid with gradient elution, lfow rate was 1.0 mL·min-1, wavelength was 367 nm, and the temperature of column was 30℃.Result: Scopoletin and hedyotiscone B's curves were linear in the ranges of 0.34 μg·mL-1 ~ 5.40 μg·mL-1and 0.32 μg·mL-1 ~5.12 μg·mL-1, solution stability, repeatability and recovery were good. The contents of two coumarins constituents of Jiangxi were higher than in other three areas, and the lowest contents came from Guangxi which did not contain hedyotiscone B.Conclusion: The method is simple, accurate, and can be used for detection of the contents of scopoletin and hedyotiscone B in H. diffusa from different areas.%目的:比较不同产地白花蛇舌草中两种香豆素类成分东莨菪亭和耳草酮B的含量,为完善白花蛇舌草药材质量标准提供参考依据.方法:采用Agilent Eclipse SB-C18色谱柱(5 μm,4.6× 250 mm),以甲醇-0.05%甲酸水为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长367 nm,柱温30℃.结果:东莨菪亭和耳草酮B分别在0.34μg·mL-1~ 5.40μg·mL-1和0.32μg·mL-1~5.12μg · mL-1范围内线性关系良好,溶液稳定性、重复性及加样回收率良好;江西产药材中两种香豆素类成分的含量明显高于湖北、安徽、广西其它三个产地的药材,而产自广西的药材含量最低且没有检出耳草酮B.结论:本方法简便,准确度高,可用于不同产地白花蛇舌草药材中东莨菪亭和耳草酮B含量的比较测定.
