Objective:To establish a UPLC-MS/MS method for the simultaneous determination of nine vitamins in four kinds of suspension cells of Xuelian,and analyze and investigate the difference of vitamin ingredients in samples from different suspension cells of Xuelian.Methods:The determination was performed on the ACQUITY UPLC BEH C18 (100 mm ×2.1 mm,1.8 μm);the 10 mmol ·L-1 ammonium acetate (A) and acetonitrile (B) in gradient elution as mobile phase.The flow rate was 0.5 mL · min-1.The column temperature was 40 ℃.The MS instrument was equipped with ESI-ion source.Gaining the extracted ion chromatograms.Results:Nine vitamins had a good linear relationship,and the r values were more than 0.999 0.The RSD values of the precision,repeatability and stability were less than 4.28％.the recoveries were 97.6％-103.4％,the RSD was 1.83％-3.12％.There were no significant differences in the composition and content of nine vitamins in the suspension cells of different Xuelian.Conclusion:The method is simple,accurate and reproducible.It is suitable for the determination of water-soluble vitamins in the suspension cells of Xuelian.%目的:建立超高效液相色谱三重四级杆串联质谱法(UPLC-MS/MS)同时测定4种雪莲培养物中9个水溶性维生素类成分的方法,分析比较不同雪莲悬浮细胞中维生素种类和含量差异.方法:采用ACQUITY UPLC BEHC18色谱柱(100mmx2.1 mm,1.8 μm),以10 mmol·L-乙酸铵(A)-乙腈(B)溶液为流动相,梯度洗脱,体积流量为0.5 mL·min-1,柱温为40℃;采用电喷雾离子源、正离子检测方式,得到相应的提取离子流图,以峰面积进行定量.结果:9个维生素类成分具有良好的线性关系,r值均大于0.999 0,方法精密度、重复性和稳定性的RSD值均小于4.28％,加样回收率在97.6％ ～ 103.4％,RSD在1.83％ ～ 3.12％.不同雪莲悬浮细胞中9个维生素类成分在组成和含量上无明显差异.结论:该方法操作简便、准确、重复性好,适用于雪莲悬浮细胞中水溶性维生素的测定.
