目的:通过人的黑色素细胞实验研究珍珠水解液的美白作用机理.方法:NaOH法测定黑色素细胞黑色素含量,实时荧光定量PCR法(RT-PCR)测定珍珠水解液对影响黑色素合成的相关基因酪氨酸酶基因(TYR)、酪氨酸酶相关蛋白1 (TRP1)及影响黑色素转运的相关基因小眼畸形相关转录因子(MITF).结果:浓度为0.25%、0.5%、0.75%的珍珠水解液可以抑制黑色素细胞的黑色素合成,3种浓度下的珍珠水解液均可抑制相关基因TRP1mRNA的表达,0.25%浓度下可抑制TYR、TRP1及MITF的mRNA的表达.结论:珍珠水解液主要通过抑制黑色素合成过程中TRP1 mRNA的表达,从而抑制黑色素合成而发挥美白功效.%Objective:To study skin-whitening mechanism of Margarita liquid by in vitro melanoma cells tests.Methods:The skin-whitening activities were evaluated by measuring human melanocytes melanin synthesis inhibition activity,and Real-time PCR (RT-PCR)method was further used to evaluate the expression of microphthalmia-associated transcription factor (MITF),tyrosinase-related protein 1 (TRP1) and Tyrosinase(TYR)mRNA.Results:0.25%,0.5% and 0.75% concentration of Margarita liquid could significantly decreased human melanocytes melanin synthesis,all the three concentrations of Margarita liquid could decrease the expression of TRP1 mRNA,and 0.25% concentration of Margarita liquid could decrease the expression of TYR、TRP1 and MITF mRNA.Conclusion:Margarita liquid was detected to have skin-whitening activity through in vitro Human melanoma cells tests,it could be used as a skin-whitening cosmetic additive.
