幽门螺杆菌感染激活人蛋白相互作用子网的构建

     

摘要

目的 从整体水平探索幽门螺杆菌(Hp)感染后宿主的蛋白相互作用网络,挖掘其感染宿主细胞和导致宿主细胞病变的关键基因,为Hp感染的机制提供新的线索,可以为细茵-宿主联合靶向治疗提供基础.方法 利用Hp感染的宿主细胞表达谱,基于激活子网算法构建了Hp感染激活子网,并对子网中的基因进行基因本体(GeneOntologY,GO)分类.结果 通过GO分类发现,子网中主要涉及宿主免疫反应的基因、细胞骨架相关基因和细胞增殖相关基因.结论 幽门螺杆茵侵染宿主主要引发宿主的免疫反应、细茵进入宿主细胞的反应和宿主增殖与病变的反应.%Objective To study the protein-protein interaction network of the host after Helicobacter pylori( Hp ) infection on a global level, exploit key infectious and cytopathic genes, and to provide a new clue to the mechanism of Hp infection and data for bacterium-host combination targeting treatment. Methods In this paper, we used a host cell expression profile from Hp infection, built an active subnetwork for Hp infection based on the active subnetwork algorithm, and made gene ontology( GO ) classification from active subnetwork. Results Through GO classification, the active subnetwork was found to be mainly related to host immunoreactive genes, cytoskeleton associated genes and cell proliferation genes. Conclusion Hp infection mainly induces the host immune response, reaction on bacterial entrance and host cell proliferation and pathological changes.
