抗人前列腺干细胞抗原单克隆抗体的制备及筛选

摘要

Objective To prepare and select monoclonal antibodies ( mAb) against human prostate stem cell antigens (PSCA) in cells. Methods BALB/c mice were immunized with recombinant human PSCA . Anti-PSCA mAbs were obtained by hybridoma technique , tested by ELISA , Western blotting and flow cytometry. Results Six strains of anti -PSCA mAbs were acquired, three of which proved to be able to bind PSCA in cells after all tests . Conclusion Acquisition of an-ti-PSCA mAbs able to bind PSCA on the cell surface will facilitate further studies and applications .%目的 获得能够结合细胞表面前列腺干细胞抗原(prostate stem cell antigen,PSCA)的抗人PSCA单克隆抗体.方法 以重组人PSCA蛋白为免疫原,通过杂交瘤技术制备抗人PSCA单克隆抗体,并进行ELISA、Western印迹、流式细胞术等实验筛选.结果 获得6株抗人PSCA单克隆抗体,经过筛选最终得到3株能够结合细胞表面PSCA的单克隆抗体.结论 获得具有结合细胞表面PSCA活性的单克隆抗体,为进一步研究和应用奠定了基础.
