黄芪皂苷Ⅳ对大鼠脑缺血/再灌注后海马血管生成的影响

     

摘要

Objective To investigate the effert of astragaloside Ⅳ on vascular endothelial growth factor ( VECF ) and microvessel density ( MVD ) in hippocampal region after cerebral ischemia-reperfusion ( CIR )in rats . Methods Rats were randomly divided into sham operation group, model group, nimodipine group and Astragaloside Ⅳ group. 72 hours after reperfusion,neurological function score were observed, MVD was evaluated by immunohistochemistry with the specific antihody CD34 . The expressions of VEGF proteins was ohserved bv using immunohistochemical and Western blot. Results Compared with model and nimodipine group, the neurological scores of astragaloside Ⅳ group was significantly decreased. Compared with model and nimodipine group. The expression of VEGF protein and MVD were significantly increased in astragaloside Ⅳ group. Conclusion Astragaloside Ⅳ could improve the neurological function in rats after cerebral ischemiareperfusion , which may be related to up-regulating VEGF protein and MVD in hippocampal region.%目的 探讨黄芪皂苷Ⅳ对大鼠缺血/再灌注损伤后海马中血管内皮生长因子(VEGF)和微血管密度(MVD)表达的影响.方法 实验动物随机分为假手术组、模型组、尼莫地平对照组、黄芪皂苷Ⅳ组.缺血2 h再灌注72 h后,各组进行神经功能评分,采用Western blot及免疫组化方法检测VEGF蛋白表达,免疫组化法进行CD34染色计数MVD.结果 黄芪皂苷Ⅳ组神经功能损害评分低于模型组和尼莫地平对照组(P<0.05);与尼莫地平对照组和模型组比较,黄芪皂苷Ⅳ组可明显上调VEGF蛋白表达及MVD(P<0.05).结论 黄芪皂苷Ⅳ对大鼠脑缺血/再灌注有神经保护作用,其作用机制可能与促进海马VEGF蛋白表达,增加MVD有关.
