Smad4蛋白在胰腺癌中的表达研究

         

摘要

Objective To detect the expression of tumor suppressor gene Smad4 in pancreatic carcinoma and carcinoma-adjacent tissue by tissue microarray, and identify protein Smad4 expression in pancreatic cancer and its clinical significance.Methods We determined Smad4 expression in 162 human pancreatic neoplasm and in 162 paracancerous specimens, which were proven by surgery or autopsy, using immunohisto-chemistry and tissue microarray.Results The expression of Smad4 in pancreatic carcinoma and carcinoma-adjacent tissue were located in the cytoplasm and nucleus.The expression level of the cytoplasm in carcinoma-adjacent tissue was significantly higher than that of pancreatic carcinoma tissue ( P < 0.05 ), but the expression level of the nucleus in pancreatic carcinoma tissue was significantly higher than that of carcinoma-adjacent tissue( P < 0.05 ).There was no correlation between the expression level of Smad4 and tumor size and pathological grade( P >0.05 ).But there was negative correlation between the dyeing index of Smad4 in the lymph node metastasist P <0.05 ).Conclusion The Smad4 gene is related to the occurrence and development of pancreatic carcinoma,and Smad4 expression level is related to lymph node metastasis.The detection of gene Smad4 is expected to be a molecular index of early diagnosis of pancreatic carcinoma.%目的 运用组织芯片技术,检测Smad4在胰腺癌旁组织及癌旁组织中蛋白水平的表达,明确Smad4蛋白在胰腺癌中的表达及临床意义.方法 选取经手术、病理证实的胰腺癌及癌旁组织各162例,构建胰腺癌组织芯片,采用免疫组化染色法检测胰腺癌及癌旁组织中Smad4蛋白的表达.结果 Smad4在胰腺癌和癌旁组织中的表达定位于细胞质和细胞核,在癌旁组织细胞质内显著高于癌组织(P<0.05),而在癌组织细胞核内显著高于癌旁组织(P<0.05).胰腺癌及癌旁组织Smad4与肿瘤大小、病理分级均无相关性(均P>0.05);而癌旁组织中胞核Smad4的染色指数与淋巴结转移呈负相关(P<0.05).结论 Smad4与胰腺癌的发生、发展有关,且与淋巴结转移与否相关,该基因有望成为胰腺癌早期诊断的分子指标.
